Determination of formaldehyde in sea fishes by high performance liquid chromatography

    摘要:

    建立了海捕鱼肌肉组织中甲醛的高效液相色谱测定方法。样品用蒸馏水浸泡30 min后,在磷酸介质中加热,经水蒸气蒸馏,甲醛冷凝后被水吸收。取收集液经过3.0 g/L的2,4–二硝基苯肼溶液在60 ℃水浴条件下衍生化20 min,快速冷却后高效液相–紫外检测法测定。结果表明,甲醛在0.20~15.0 mg/L质量浓度范围内,线性相关系数均大于0999 9;在 2.0~20.0 mg/kg的添加范围内的平均回收率为92.5%~98.0%,相对标准偏差均小于5.0%;方法检出限为0.20 mg/kg。本方法基质干扰小,目标物出峰好,重复稳定性高。

    Abstract:

    High performance liquid chromatography (HPLC) was applied to the determination of formaldehyde in muscle tissues of sea fishes. The samples were soaked with distilled water for 30 min, and then heated in phosphoric medium. Formaldehyde was absorbed by water using steam distillation method. The collection was taken and derived with 2, 4-dinitrophenylhydrazine under the condition of 60 ℃ water bath for 20 min.The derivative was determined by HPLC after rapid cooling. In the range of 0.20-15.0 mg/L, linear correlation coefficients were all above 0.999 9. And in the range of 2.0-20.0 mg/kg, the mean fortification recoveries were between 92.5% and 98.0%, The RSDs were all less than 5.0%, and the detection limit was 0.20 mg/kg. The method had little matrix interference, proper target peak, and higher repeatability and stability.
