    本实验采用体长10.34±1.33 cm、体重19.0±3 g的活鱼,并用聚乙烯袋密闭充氧运输方式,在3种不同运输时间(4 h、8 h、12 h)下,探究高体鰤应激、代谢、抗氧化和免疫的影响。结果显示,随着运输时间的增加,水体中氨氮(NH4-N)浓度升高,而pH和溶解氧(DO)降低。在肝脏组织中,4 h组谷胱甘肽还原酶(GR)表达水平和丙酮酸激酶(PK)表达水平均显著高于8 h和12 h(P<0.05);12 h的组铜超氧化物歧化酶(Cu-SOD)表达水平和锰超氧化物歧化酶(Mn-SOD)表达水平均显著高于4 h和8 h (P<0.05);12 h时的丙二醛(MDA)浓度显著高于4 h和8 h(P<0.05);主要组织相容性复合物(MHC-1)在4 h、8 h、12 h三组间均存在显著差异(P<0.05)。在鳃中,8 h时的 GR的表达水平和PK的表达水平均显著高于4 h和12 h;8 h时的Cu-SOD表达水平显著高于4 h和12 h 组(P<0.05);4 h时的Mn-SOD表达水平显著低于8 h和12 h (P<0.05);12 h时的MDA浓度显著高于4 h和8 h ;12 h的MHC-1表达水平显著低于4 h和8 h 。随着时间的增加,鱼类在运输过程中会影响水质(酸碱度、氨氮、溶解氧),导致鱼体肝脏、鳃损伤。所以运输最好控制在8 h内,减少对鱼体生理功能、及免疫功能的损伤。

    Abstract:

    Live fish of 10.34±1.33 cm in length and 19.0±3 g in weight were transported in polyethylene bags for three different transport times (4 h, 8 h and 12 h) to investigate the effects on Seriola dumerili amberjack stress, metabolism, antioxidant and immunity. The results showed that ammonia and nitrogen (NH4-N) concentrations in the water column increased with increasing transport time, while pH and dissolved oxygen (DO) decreased. In liver tissues, the expression levels of glutathione reductase (GR) and pyruvate kinase (PK) were significantly higher in the 4 h group than in the 8 h and 12 h groups (P < 0.05); the expression levels of copper superoxide dismutase (Cu-SOD) and manganese superoxide dismutase (Mn-SOD) were significantly higher in the 12 h group than in the 4 h and 8 h groups (P < 0.05); the 12 h group Malondialdehyde (MDA) concentration was significantly higher in the 12 h group than in the 4 h and 8 h groups (P < 0.05); major histocompatibility complex (MHC-1) was significantly different between the 4 h, 8 h and 12 h groups (P < 0.05). In the gills, the expression levels of GR and PK were significantly higher in the 8 h group than in the 4 h and 12 h groups; the expression levels of Cu-SOD were significantly higher in the 8 h group than in the 4 h and 12 h groups (P < 0.05); the expression levels of Mn-SOD were significantly lower in the 4 h group than in the 8 h and 12 h groups (P < 0.05); the MDA concentration was significantly higher in the 12 h group than in the 4 h and 8 h groups; the MHC-1 The expression level of MHC-1 in the 12 h group was significantly lower than that in the 4 h and 8 h groups. As the time increases, the fish will affect the water quality (pH, ammonia and nitrogen, dissolved oxygen) during transportation, leading to damage to the liver and gills of the fish. Therefore, it is best to control the transportation within 8 h to reduce the damage to the physiological function and immune function of fish.
